چکیده
By L. A. MOUNTER AND WASFI ATIYEH T HE EXISTENCE of considerable proteolvtic activities associated with leukocytes has previously been reported, but little characterization of the enzymes has been carried out.13 Experiments with leukocyte preparations using newer enzyme inhibitors are very limited and it seemed probable that improved characterization might result from studies of this type. It was also hoped that the studies might indicate whether leukocyte protease activity could be sufficient to contribute appreciably to the breakdown of damaged tissues invaded by a high concentration of white cells. The present report describes the results of preliminary studies indicating the existence of at least three intracellular proteolytic enzymes in human leukocytes.

Human leukocyte cathepsin G and elastase specifically suppress thrombin-induced prostacyclin production in human endothelial cells.
Polymorphonuclear leukocytes (PMN) when activated release products that can potentially injure endothelial cells or alter endothelial function. Exposure of cultured human umbilical vein endothelial cells to cathepsin G and elastase isolated from human PMN at concentrations reached in vivo (100 ng/mL to 10 micrograms/mL) selectively inhibited thrombin-induced prostacyclin production and the thro...
متن کاملSubcellular Localization and Heterogeneity of Neutral Proteases in Neutrophilic Polymorphonuclear Leukocytes
The subcellular localization of elastase and of neutral proteases hydrolyzing histone and casein was determined in human and rabbit polymorphonuclear leukocytes using fractionation by isopycnic centrifugation. Granule-rich fractions obtained by this technique were extracted and analyzed by acrylamide gel electrophoresis, and proteolytic activity on the gels was demonstrated by staining with eit...
متن کاملConsumption of antioxidant dietary agents, curcumin and vitamin C, protects cellular DNA from gamma-radiation
Background: Exposure to ionizing radiation results in genotoxicity and the unrepaired lesions in cellular DNA results in cell cycle arrest, reproductive death, interphase death, division delay, chromosome aberrations, mutations, etc. leading to the intensive destruction of cells and violation of their proliferative capacity there by adversely affecting the mammalian system. Since ionizing radia...
متن کاملIsolation and Characterization of Thermophilic Alkaline Proteases Resistant to Sodium Dodecyl Sulfate and Ethylene Diamine Tetraacetic Acid from Bacillus sp. GUS1
Thermophilic Bacillus sp. GUS1, isolated from a soil sample obtained from citrus garden, produced at least three proteases as detected by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and zymogram analysis. The enzymes were stable in the alkaline pH range (8.0-12.0), with the optimum temperature and pH range of the proteases being 70ºC and 6.0-12.0, respectively. All th...
متن کاملEffect of famotidine on radiation induced apoptosis in human peripheral blood leukocytes
ABSTRACTBackground: Radioprotective effects of famotidine, an antagonist of H2 receptor clinically used for peptic ulcer treatment, was previously shown on radiation-induced micronuclei and chromosomal aberration in human peripheral blood lymphocytes and mouse bone marrow cells. This study was conducted to investigate radioprotective property of famotidine against radiation induced apoptosis i...
